This study analyzes the microbial communities in leachate-contaminated soil in Jalan Lipis Sanitary Landfill. The study also investigated how the heavy metal (Mn, Ni, As, Pb) concentration and pH influences the diversity and distribution of the microbial communities. This sanitary landfill received domestic waste from all over Lipis. The estimated area is about 10.22 ha and receives roughly about 51.45 tonnes of waste daily. The extraction of gDNA was done by using a GF-1 Soil sample DNA extraction kit (Vivantis Technologies, Malaysia). The bacterial 16S V3 hypervariable region was amplified using the universal bacterial primers (341F and 518R) with a partial Illumina adapter.
本研究针对惹兰立皮斯(Jalan Lipis)卫生填埋场受渗滤液污染的土壤中的微生物群落展开分析,同时探究了重金属(锰Mn、镍Ni、砷As、铅Pb)浓度与pH值对微生物群落多样性及分布格局的影响。该卫生填埋场接收来自立皮斯全域的生活垃圾,预估占地面积约10.22公顷,日均接收生活垃圾约51.45吨。实验采用GF-1土壤样本DNA提取试剂盒(马来西亚Vivantis Technologies公司)完成基因组DNA(gDNA)的提取,并使用带有部分Illumina接头的通用细菌引物(341F与518R)对细菌16S核糖体RNA V3高变区进行扩增。
