Clostridium difficile strains have been investigated for the presence of functional prophages. Following mitomycine C induction, temperate phages have been isolated. Transmission electron microscopy (TEM) has revealed member of Siphoviridae / Myoviridae family. Phage DNA has been purified following phage amplification on sensitive host and subjected to complete genome sequencing using Illumina technology. Following sequencing, double stranded-DNA (dsDNA) genomes have been assembled and annotated.
本研究针对艰难梭菌(Clostridium difficile)菌株的功能性前噬菌体存在情况展开了探究。经丝裂霉素C(mitomycine C)诱导处理后,成功分离得到温和噬菌体。借助透射电子显微镜(Transmission Electron Microscopy, TEM)观测,确认分离得到的噬菌体隶属于长尾噬菌体科(Siphoviridae)与肌尾噬菌体科(Myoviridae)。通过在敏感宿主菌中扩增噬菌体,纯化获得噬菌体DNA,并采用Illumina测序技术完成全基因组测序。测序完成后,对双链DNA(double-stranded DNA, dsDNA)基因组进行了组装与注释。
