D. nodosus WT vs pilR mutant

The aim was to determine the additional genes regulated by PilR in addition to fimA. To determine the additional genes the gene expression profile of a pilR mutant was compared to the wild-type strain. Keywords: genetic modification PilR regulated genes were determined by analysed microarray expression data from three biololgical replicates, including a dye swap. The expression data from the pilR mutant was compared to the wild-type strain. Raw data was obtained using IMAGENE, while normalisation and analysis was performed using BASE. Statistical analysis was undertaken using the Wilcoxon signed rank test.

本研究旨在探究除fimA之外，受PilR调控的其他基因。为明确该类靶基因，本研究将pilR突变株的基因表达谱与野生型菌株进行了对比分析。关键词：基因修饰（genetic modification）。本研究通过分析三次生物学重复（含染料互换实验）的微阵列表达数据，确定了受PilR调控的基因；研究中同样将pilR突变株的表达数据与野生型菌株进行了比对。原始数据通过IMAGENE软件获取，表达数据的归一化与分析则借助BASE软件完成。统计分析采用Wilcoxon符号秩检验（Wilcoxon signed rank test）。
