针对杜仲叶提取产物中绿原酸含量低、杂质复杂多样的缺点,研究首先采用取法去除杂质,提高绿原酸的含量,利用石油-乙酸乙酯苯取的办法,在不失绿原酸的情况下去除脂溶性成分,再利用乙酸乙酯在酸性条件下(pH≤2)对绿原酸进行有效富集。下一步,研究通过对分离树脂型的筛选,选定HP-20大孔树脂进行绿原酸富集产物的进一步纯化,无论是小试实验还是中试化生产,都能有效地对其中的绿原酸进行纯化。成果已经进行了产业化,和西安锐博生物科技有限公司签订了成果转让协议。
Aiming at the drawbacks of low chlorogenic acid content and complex and diverse impurities in the extract of Eucommia ulmoides leaves, this study first adopted extraction methods to remove impurities and increase the chlorogenic acid content. Specifically, petroleum ether-ethyl acetate extraction was used to remove lipophilic components without losing chlorogenic acid, followed by effective enrichment of chlorogenic acid with ethyl acetate under acidic conditions (pH ≤ 2). Next, through screening of separation resin types, HP-20 macroporous resin was selected for further purification of the chlorogenic acid-enriched product. This method can effectively purify chlorogenic acid in the product, whether in small-scale laboratory experiments or pilot-scale production. The research results have been industrialized, and a technology transfer agreement has been signed with Xi'an Ruibo Biotechnology Co., Ltd.
