Whole genome amplification (WGA) has been used to amplify DNA from low biomass samples for downstream omics analysis with the risks of introducing biases and skewing community profiles. In this study, two WGA methods, Multiple Displacement Amplification (MDA) and Multiple Annealing and Looping-Based Amplification Cycles (MALBAC), were evaluated with three contrasting soils and by amplicon sequencing of 16S rRNA genes. The performance of these two WGA methods was compared in terms of the reconstruction of bacterial communities.
全基因组扩增(Whole genome amplification, WGA)常被用于从低生物量样本中扩增DNA以开展下游组学分析,但该技术存在引入扩增偏差、扭曲微生物群落结构的风险。本研究选取三种特性迥异的土壤作为实验材料,对两种全基因组扩增方法——多重置换扩增(Multiple Displacement Amplification, MDA)与多重退火和成环循环扩增(Multiple Annealing and Looping-Based Amplification Cycles, MALBAC)进行评估,通过16S核糖体RNA基因(16S rRNA genes)扩增子测序,对比二者在细菌群落组成重构方面的性能。
