The study was designed to expolre the differential gene expression after peroxiredoxin2 (Prdx2) downregulation by siRNAs transfection in human trophoblast cell line HTR8 compared with negative control. siRNA targeting the human Prdx2 or negative control siRNA were transfected into HTR8 cells. Cells were cultured for 48 h after transfection before they were harvested. Three independent experiments were performed for each sample. The Agilent SurePrint G3 Human Gene Expression v3 8x60K Microarray was used to detect gene expression.
本研究旨在探究人滋养层细胞系HTR8中,经小干扰RNA(small interfering RNA, siRNA)转染下调过氧化物还原蛋白2(peroxiredoxin2, Prdx2)的表达后,与阴性对照组相比的差异基因表达情况。将靶向人Prdx2的siRNA或阴性对照siRNA转染至HTR8细胞内。转染完成后,将细胞培养48小时再进行收集。每个样本均开展三次独立重复实验。采用安捷伦SurePrint G3人基因表达v3 8x60K微阵列芯片检测基因表达水平。
