We applied scRNA-seq to map organism-level cell landscapes at multiple life stages for mice, zebrafish, and Drosophila. By integrating the comprehensive dataset of more than 2.6 million single cells, we constructed a cross-species cell landscape and identified signatures and common pathways that changed throughout the lifespan. We identified structural inflammation and mitochondrial dysfunction as the most common hallmarks of organism ageing, and found that pharmacological activation of mitochondrial metabolism alleviated ageing phenotypes in mice. Meanwhile, the cross-species cell landscape with other published data sets were stored in an integrated online portal Cell Landscape (http://bis.zju.edu.cn/cellatlas/).

本研究采用单细胞RNA测序(scRNA-seq)技术,对小鼠、斑马鱼及果蝇(Drosophila)的多个生命阶段的生物体水平细胞图谱进行绘制。通过整合超260万个单细胞的综合性数据集,我们构建了跨物种细胞图谱,并鉴定出贯穿整个生命周期的细胞特征与共通通路。我们将结构性炎症与线粒体功能障碍鉴定为生物体衰老最常见的标志性特征,同时发现线粒体代谢的药理学激活可缓解小鼠的衰老表型。与此同时,本跨物种细胞图谱及其他已发表数据集均存储于整合在线平台“细胞图谱(Cell Landscape)”(http://bis.zju.edu.cn/cellatlas/)中。
