Dissecting Cell Composition and Drug Sensitivity in Human Adenoid Cystic Carcinomas (ACCs)

The study reports the results of single-cell RNA sequencing (scRNA-seq), RNA sequencing (RNA-seq), and whole-exome sequencing (WES) experiments performed on malignant cells isolated by flow cytometry from seven patient-derived xenograft (PDX) lines established from human Adenoid Cystic Carcinomas (ACCs). Of these seven lines, five are characterized by well differentiated histology (ACCX5M1, ACCX6, ACCX14, ACCX22, SGTX6) and two by solid histology (ACCX9, ACCX11). Analysis by scRNA-seq of the well-differentiated PDX line ACCX22 led to identification of cell-surface markers (CD49f, KIT) that enabled the differential purification of the two major cellular components contained in well-differentiated ACCs: myoepithelial-like (CD49f-high/KIT-negative) and ductal-like (CD49f-low/KIT-positive)... 本研究报道了针对7株由人类腺样囊性癌(Adenoid Cystic Carcinomas, ACCs)构建的患者来源异种移植模型(patient-derived xenograft, PDX)中,经流式细胞术分离的恶性细胞所开展的单细胞RNA测序(single-cell RNA sequencing, scRNA-seq)、RNA测序(RNA sequencing, RNA-seq)及全外显子组测序(whole-exome sequencing, WES)实验结果。该7株模型中,5株具备高分化组织学特征(ACCX5M1、ACCX6、ACCX14、ACCX22、SGTX6),剩余2株为实体型组织学类型(ACCX9、ACCX11)。针对高分化PDX模型株ACCX22的单细胞RNA测序分析,成功鉴定出细胞表面标志物CD49f与KIT,借助二者可实现高分化腺样囊性癌中两种主要细胞组分的差异化纯化:肌上皮样细胞(CD49f高表达/KIT阴性)及导管样细胞(CD49f低表达/KIT阳性)……(详细内容参见dbGaP研究页面)
