To investigate the cell-cell interactions of intergeneric bacterial species, the study analyzed the transcriptomic profiling of coaggregated Enterococcus faecalis (Ef) and Fusobacterium nucleatum subsp. polymorphum (Fnp). Ef and Fnp were coaggregated in the coaggregation buffer. By using RNA-Seq analysis, gene regulation responses between mono-species and coaggregated species were compared, and differentially expressed genes were screened using DESeq2. Real-time quantitative polymepolymerase chain reaction was carried out to verify the results of DEGs. Bioinformatics analysis concerning DEGs was performed through KEGG pathway, GO functional enrichment and protein protein interaction network.

为探究跨属细菌的细胞间互作机制,本研究对共聚集状态下的粪肠球菌(Enterococcus faecalis, Ef)与具核梭杆菌多形亚种(Fusobacterium nucleatum subsp. polymorphum, Fnp)的转录组谱进行了分析。两菌种于共聚集缓冲液中完成共聚集;通过RNA测序(RNA-Seq)分析,对比单菌种培养与共聚集培养菌种的基因调控响应,并借助DESeq2筛选差异表达基因(DEGs);采用实时定量聚合酶链式反应对差异表达基因的测序结果进行验证;针对差异表达基因的生物信息学分析通过京都基因与基因组百科全书(KEGG)通路分析、基因本体(GO)功能富集分析及蛋白质-蛋白质相互作用网络分析完成。
