This dataset presents the molecular characterization of Human Respiratory Syncytial Virus group A (HRSV-A) genotypes circulating along the coast of Peru between 2009 and 2020. Using a retrospective analysis of 1,694 nasopharyngeal swab samples from pediatric patients hospitalized with acute respiratory infections, 53 sequences of the G glycoprotein gene were analyzed. Phylogenetic reconstructions using Bayesian inference and Maximum Likelihood methods identified the co-circulation of the NA1 and ON1 genotypes, with a newly proposed Peruvian lineage ON1[1-6] exhibiting three exclusive point mutations (S250F, S267L, and T293I). These mutations reduced O-glycosylation potential and may alter antigenic properties of the virus. Findings suggest that the ON1 genotype may have originated in South America, possibly Peru, earlier than previously reported, and emphasize the need for continuous genomic surveillance.
