WXS of mouse DNA using a human enrichment kit

Sequence data (RNA/DNA) of patient derived Xenografts (PDX)not only contains reads of human (tumor) origin but also of mouse (host) origin. We developed XenofilteR, a tool to computationally dissect human and mouse sequence reads from xenograft sequence data. One of the validation sets for the tool includes sequence data of mouse DNA enriched with a human enrichment kit. This to show that also in WXS data murine sequence reads are present and hybridize with the baits designed based on the human genome.

患者来源异种移植瘤（patient-derived xenografts, PDX）的RNA/DNA序列数据，不仅包含人类（肿瘤）来源的测序读段，同时也包含小鼠（宿主）来源的测序读段。我们开发了XenofilteR工具，可通过计算手段从异种移植瘤序列数据中分离人类与小鼠来源的测序读段。该工具的一组验证集包含了使用人类富集试剂盒处理的小鼠DNA序列数据，此举旨在证明，在全外显子组测序（Whole Exome Sequencing, WXS）数据中同样存在小鼠来源的测序读段，且这些读段会与基于人类基因组设计的捕获探针发生杂交。
