Cultured organotypic cerebellar slices were exposed for different time points with either prions (RML) versus non-infectious brain homogenate (NBH) or ligands to the globular domain of the prion protein (POM1) vs IgG We used microarrays to characterize the temporal response of two inducers prion protein dependent cell death: COCS were exposed to RML prions or Non-infectious brain homogenate (NBH) and harvested after 14d, 25d, 38d and 45d (4 replicates each condition). COCS were exposed to POM1 or IgG and harvested after 8h, 1d, 3d, 7d, 10d (4 replicates each condition).
将培养的小脑器官型切片分为两类实验体系,分别以不同时长暴露于对应的处理与对照因素:其一为RML株朊病毒(prions)与非感染性脑匀浆(non-infectious brain homogenate, NBH)对比组;其二为靶向朊蛋白(prion protein)球状结构域的配体POM1与免疫球蛋白G(IgG)对比组。
为表征两种诱导朊蛋白依赖性细胞死亡的诱导剂的时序应答特征,本研究采用微阵列(microarrays)技术开展检测:小脑器官型切片培养(cultured organotypic cerebellar slices, COCS)经RML朊病毒或非感染性脑匀浆(NBH)暴露后,分别于14天、25天、38天、45天收获样本,每组设置4次生物学重复。
COCS经POM1或IgG暴露后,分别于8小时、1天、3天、7天、10天收获样本,每组设置4次生物学重复。
