Fu Huang ointment promotes wound healing via the LPAR1/ EDN1 axis based on transcriptome analysis

Clinically, topical Fu Huang ointment has achieved good results in the treatment of diabetic ulcers, but its specific mechanisms of action are unclear. Firstly, diabetic ulcers animal model was established by using db/db mice combined with surgical resection. Secondly, after 7 days of administration, the exact therapeutic effect of Fu Huang ointment on diabetic ulcers was evaluated by observing the wound healing rate and detected the angiogenesis, granulation tissue formation, and inflammatory response in local wound tissue. Then, total RNA Sequencing was performed on the tissue samples of diabetic ulcers to screen core target genes to determine the regulatory mechanism by which Fu Huang ointment exerts its therapeutic effect. Finally, further verification was based on animal experiments.

临床研究显示,外用复黄膏(Fu Huang ointment)在糖尿病溃疡治疗中已取得良好疗效,但具体作用机制尚不明确。本研究首先采用db/db小鼠联合外科切除法构建糖尿病溃疡动物模型;其次,给药7天后,通过观察创面愈合率、检测局部创面组织的血管生成、肉芽组织形成及炎症反应,评估复黄膏对糖尿病溃疡的确切治疗效果;随后,对糖尿病溃疡组织样本开展总RNA测序(total RNA Sequencing),筛选核心靶基因,以明确复黄膏发挥治疗作用的调控机制;最后,通过动物实验完成进一步验证。
