RNA transcriptome sequencing (RNA-Seq) and RNA reverse transcription-associated trap sequencing (RAT-Seq) were usded to map long noncoding RNA (lncRNAs) in reprogramming of fibroblasts into pluripotency Overall design: Fibroblasts (FIB) and induced pluripotent stem cells (iPSCs) were collected in the process of reprogramming. RNA-Seq was used to identify lncRNAs that are differentially-expressed in reprogramming. RAT-Seq was utilized to generate the database to map the regulatory element network of lncRNA candidate Osclr8 (NONMMUT043505, Platr10).

本研究采用RNA转录组测序(RNA transcriptome sequencing,简称RNA-Seq)与RNA反转录关联捕获测序(RNA reverse transcription-associated trap sequencing,简称RAT-Seq),对成纤维细胞重编程为多能干细胞过程中的长链非编码RNA(long noncoding RNA,简称lncRNAs)进行定位分析。总体实验设计:在成纤维细胞重编程进程中收集成纤维细胞(FIB)与诱导多能干细胞(induced pluripotent stem cells,简称iPSCs);借助RNA-Seq筛选重编程过程中差异表达的长链非编码RNA;利用RAT-Seq构建数据库,以绘制候选长链非编码RNA Osclr8(NONMMUT043505,Platr10)的调控元件网络。
