We developed human CISH-knockout (CISH-/-) NK cells using an induced pluripotent stem cell-derived NK cell (iPSC-NK cell) platform. And compare transcriptomes of CISH-/- iPSC NK cells with wild type iPSC NK cells using RNA seq. RNA seq data suggested that genes involved in the JAK-STAT signaling pathway and lymphocyte activations were significantly activated in CISH-/- iPSC NK cells. mRNA profiles of wild type and CISH knockout iPSC NK cells were generated by deep sequencing, in dulpicate, using Illumina HiSeq 2000.

本研究利用诱导多能干细胞衍生NK细胞(induced pluripotent stem cell-derived NK cell)平台,构建了人源CISH基因纯合敲除(CISH-/-)NK细胞。随后通过RNA测序(RNA-seq)对比CISH-/- iPSC-NK细胞与野生型iPSC-NK细胞的转录组。RNA测序数据显示,JAK-STAT信号通路(JAK-STAT signaling pathway)及淋巴细胞活化相关基因在CISH-/- iPSC-NK细胞中显著激活。本研究采用Illumina HiSeq 2000平台进行双重复深度测序,获得了野生型与CISH基因敲除iPSC-NK细胞的mRNA表达谱。
