Skin homeostasis is guided by spatiotemporal regulation of gene expression, establishing each stage of keratinocyte differentiation. The current study presents transcriptomic and chromatin profiling for the proliferating (basal) and differentiated (suprabasal) cell populations derived from neonate mice (p0-p2) skin epidermis. This multi-omic approach will enable idenitification of cell specific epidermal cross-talk central to the equlibirum. Overall design: Transcriptomic profiling for adeno-virus transduced (either Ad-GFP or Ad-Dlx3) primary cells cultured from transgenic Dlx3 wild-type and knock-out pups (p0-p1)
皮肤稳态由基因表达的时空调控所介导,进而确立角质形成细胞(keratinocyte)分化的各个阶段。本研究针对新生小鼠(出生后0~2日龄,p0-p2)皮肤表皮来源的增殖性(基底层,basal)与分化性(棘上层,suprabasal)细胞群体,开展了转录组谱分析(transcriptomic profiling)与染色质谱分析(chromatin profiling)。该多组学方法将助力鉴定表皮细胞特异性互作,这类互作是维持皮肤稳态平衡的核心要素。总体实验设计:对源自转基因Dlx3野生型与基因敲除(knock-out)幼鼠(出生后0~1日龄,p0-p1)的原代细胞,分别经Ad-GFP或Ad-Dlx3腺病毒(adeno-virus)转导后,进行转录组谱分析。
