Clinical use of triptolide (TP) is restricted due to severe toxicity. This study assessed the protective effect of crocin (CR) as a natural antioxidant against TP-induced toxicity. Mouse Liver tissues were selected from the Control, MTP, and MTP+CR groups for transcriptome studies. Overall design: The mice were randomly placed in the following 3 groups (n=4): A. Normal saline (NC); B. 300μg/kg Moderate-dose TP (MTP); C. 300μg/kg Moderate-dose TP + 100 mg/kg CR (MTP+CR); The mice were administrated the treatments daily via oral gavage for 7 days at a volume of 0.2 ml/10 g per dose.
由于雷公藤内酯醇(triptolide, TP)具有严重毒性,其临床应用受到限制。本研究评估了作为天然抗氧化剂的番红花素(crocin, CR)对TP诱导毒性的保护作用。本研究从对照组、中剂量雷公藤内酯醇组(MTP)与中剂量雷公藤内酯醇联合番红花素组(MTP+CR)中选取小鼠肝脏组织开展转录组学研究。
实验整体设计如下:将小鼠随机分为3组(每组n=4):A组:生理盐水(NC);B组:300μg/kg中剂量雷公藤内酯醇(MTP);C组:300μg/kg中剂量雷公藤内酯醇+100mg/kg番红花素(MTP+CR);所有小鼠每日以0.2ml/10g体重的给药体积经口灌胃给予对应处理,连续给药7天。
