We used single-cell sequencing to detect tumor cell types in mouse subcutaneous MC38 colorectal cancer Overall design: Solid tumors from mice were excised and mechanically dissociated into single cells using Tumor Dissociation Kit (SHBio). Single cells were captured in the 10 × Genomics Chromium Single Cell Solution, and RNA-seq libraries were prepared following the manufacturer’s protocol (10 × Genomics). The libraries were subjected to high-throughput sequencing on an Illumina Hiseq X Ten PE150 platform, and 150-bp paired-end reads were generated.

本研究采用单细胞测序技术，检测小鼠皮下MC38结直肠癌模型中的肿瘤细胞类型。
实验设计概述：将小鼠实体瘤组织切除后，使用肿瘤组织解离试剂盒（Tumor Dissociation Kit，SHBio）通过机械解离法制备为单细胞悬液。采用10× Genomics Chromium单细胞系统捕获单细胞，并按照厂商提供的官方实验流程构建RNA测序文库。将构建完成的文库置于Illumina Hiseq X Ten PE150平台开展高通量测序，最终获得150 bp双端测序读段。
