Trangenic mice with brain-targeted deletion of one allele of p53 and one allele ot Pten were irradiated with particulate radiation. Radiation induced gliomas were analyzed to identify copy number variations DNA extracted from one non-irradiated, non-tumor bearing mouse brain tissue and tumor tissue from seven mice with radiation-induced gliomas. Thus, a single DNA extraction from normal mouse brain was produced, labeled with Cy3 and co-hybridized with each of the 7 irradiated samples.
将经脑靶向单等位基因p53与单等位基因PTEN敲除的转基因(Transgenic)小鼠施以粒子辐射照射。针对辐射诱导产生的胶质瘤开展分析,以鉴定拷贝数变异(Copy Number Variation)。实验提取了1只未接受照射且无肿瘤的小鼠脑组织,以及7只辐射诱导胶质瘤小鼠的肿瘤组织中的DNA。由此,我们制备了一份来自正常小鼠脑组织的DNA提取物,采用Cy3进行标记,并将该标记产物与7个辐射样本分别进行共杂交。
