In order to examine the link between Nprl3 regulation of cellular metabolism and erythropoiesis, we performed RNA-seq and identified gene expression changes in Nprl3 +/- and Nprl3 -/- mouse fetal liver erythroblasts compared with wild type Nprl3 +/+ erythroblasts. Overall design: Poly(A)+ mRNA was analysed from four Nprl3 +/+ fetal livers (2 male, 2 female), two Nprl3 +/- fetal livers (2 male) and two Nprl3 -/- fetal livers (1 male, 1 female).
为探究Nprl3对细胞代谢的调控与红细胞生成之间的关联,我们开展了RNA测序(RNA-seq)实验,对比分析了Nprl3 +/-、Nprl3 -/- 小鼠胎肝成红细胞与野生型Nprl3 +/+ 成红细胞的基因表达变化。
整体实验设计:本研究对4份Nprl3 +/+ 胎肝样本(2份雄性、2份雌性)、2份Nprl3 +/- 胎肝样本(均为雄性)以及2份Nprl3 -/- 胎肝样本(1份雄性、1份雌性)中的聚腺苷酸化mRNA(Poly(A)+ mRNA)进行了测序分析。
