Stat2敲除小鼠在免疫反应会表现出缺陷,增加对病毒感染的易感性。利用 CRISPR 技术剪切目标基因 Stat2 的蛋白编码区,小鼠受精卵细胞通过 NHEJ 的DNA 修复 ,在蛋白编码区产生移码突变, 使得 Stat2 蛋白失活,从而实现 Stat2 基因敲除的目的
Stat2 knockout mice exhibit immune response defects and increased susceptibility to viral infections. The protein-coding region of the target gene Stat2 was cleaved via CRISPR technology. Mouse fertilized eggs underwent NHEJ-mediated DNA repair, which induced frameshift mutations within the protein-coding region, leading to the inactivation of the Stat2 protein and ultimately achieving Stat2 gene knockout.
