MLL-AF9 driven AML cells regress when LIN28B is ectopically expressed.RNA-seq was performed to measure LIN28B expression and associated changes in gene expression MLL-AF9 virus was used to induce AML from widtype or tet-on LIN28B rogenitors. After leukemia engraftment, LIN28B expression was induced in experimental mice by feeding Doxycycline chow. AML were harvested 6 days later to assess gene expression.
当LIN28B(LIN28B)异位表达时,由MLL-AF9融合基因驱动的急性髓系白血病(acute myeloid leukemia,AML)细胞会发生消退。本研究采用RNA测序(RNA-seq)技术检测LIN28B的表达水平及其相关的基因表达变化。研究人员使用MLL-AF9病毒,从野生型或四环素诱导型(tet-on)LIN28B祖细胞中诱导构建AML模型。白血病移植造模成功后,通过给实验小鼠饲喂多西环素饲料,诱导其体内LIN28B的表达。造模诱导6天后收获AML细胞以评估其基因表达情况。
