The subependymal zone (SEZ), also known as the subventricular zone (SVZ), constitutes a neurogenic niche that persists during post-natal life. To investigate the cellular diversity of this brain region during human adulthood, we characterized the complete cellular niche of the adult human SEZ by single-nucleus RNA sequencing (snRNAseq), in youth and middle-aged adults. Overall design: Single nuclei were isolated from SEZ frozen post-mortem tissue from 4 young (16-22 years) and 4 middle-aged (44-53 years) adults. DAPI positive events were collected by FACS and subjected to snRNA.

室管膜下区(subependymal zone, SEZ),又称脑室下区(subventricular zone, SVZ),是一类在出生后仍持续存在的神经发生龛(neurogenic niche)。为探究人类成年期该脑区的细胞多样性,我们采用单细胞核RNA测序(single-nucleus RNA sequencing, snRNA-seq)技术,对青年与中年成年人的成人SEZ完整细胞微环境进行了表征分析。整体实验设计:从4名青年(16~22岁)及4名中年(44~53岁)成年人的SEZ冰冻死后组织中分离单细胞核;通过荧光激活细胞分选(Fluorescence-Activated Cell Sorting, FACS)收集4',6-二脒基-2-苯基吲哚(4',6-diamidino-2-phenylindole, DAPI)阳性事件,并对其进行单细胞核RNA测序分析。
