Modulators of epithelial to mesenchymal transition (EMT) have recently emerged as novel players in the field of leukemia biology. The mechanisms by which EMT modulators contribute to leukemia pathogenesis, however, remain to be elucidated. Here we investigate the role SNAI1 - a key modulator of EMT – and its effect on chromatin state using ATAC-seq in hematopoietic cells (HPC7). Overall design: A murine hematopoietic progenitor cell line, HPC7, were transduced with a retroviral vector encoding FLAG-tagged SNAI1. Cells were harvested to generate ATAC-seq libraries for Next-Generation Sequencing.

上皮间质转化(Epithelial to Mesenchymal Transition, EMT)调控因子近期已成为白血病生物学领域的新兴研究对象。然而,EMT调控因子参与白血病发病的具体分子机制仍有待阐明。本研究针对EMT的关键调控因子SNAI1展开探究,并利用造血细胞(HPC7)中的转座酶可及性染色质测序(Assay for Transposase-Accessible Chromatin using sequencing, ATAC-seq)技术,分析其对染色质状态的影响。实验整体设计:将小鼠造血祖细胞系HPC7用携带FLAG标签SNAI1的逆转录病毒载体进行转导,收集细胞以制备用于下一代测序(Next-Generation Sequencing)的ATAC-seq文库。
