Gene expression profiling by RNA-seq of control and METTL14-knockdown HT29 human colon cancer cells

In order to define the cohort of genes whose expression is regulated by METTL14 in p53 mutant colorectal cancer, we constructed HT29 cells stably infected with lentivirus carrying control shRNA (shNC) or METTL14 shRNA (shMETTL14). Subsequently, total RNA was isolated and then performed transcriptome RNA sequence. There are four technical replicates in shNC or shMETTL14 group, respectively. Among shMETTL14 group, there existed two equal numbers of biological replicates named shMETTL14-01 an shMETTL14-02.

为了明确p53突变结直肠癌中受METTL14调控表达的基因集，我们构建了稳定感染携带对照短发卡RNA（shNC）或METTL14短发卡RNA（shMETTL14）慢病毒的HT29细胞系。随后我们提取总RNA并开展转录组RNA测序。shNC组与shMETTL14组各设置4个技术重复样本。在shMETTL14组中，包含2个生物学重复样本，分别命名为shMETTL14-01与shMETTL14-02。
