The goals of this study are to examine responses to inflammation in astrocytes from induced pluripotent stem cells derived from healthy controls and bipolar disorder patients. We examine the transcriptomic inflmmatory signature of generated astrocytes following Il1Beta exposure in BD vs. control Results: BD-patient astrocytes show a unique inflammatory response with differentially regulated genes. Human induced pluripotent stem cells (hiPSC)-derived astrocytes were treated with Il1beta and compared to vehicle treated controls from four healthy invididuals and five bipolar disorder patient individuals (run in duplicate).
本研究旨在探究健康对照者与双相情感障碍(bipolar disorder, BD)患者来源的人诱导多能干细胞(human induced pluripotent stem cells, hiPSC)分化所得星形胶质细胞(astrocytes)对炎症刺激的应答反应。本研究分析了经白细胞介素1β(Il1Beta)刺激后的双相情感障碍组与对照组星形胶质细胞的转录组炎症特征。研究结果显示,双相情感障碍患者来源的星形胶质细胞呈现独特的炎症应答模式,伴随差异调控基因的异常表达。本实验将4名健康个体与5名双相情感障碍患者来源的hiPSC分化星形胶质细胞经Il1Beta处理,并以溶剂处理组作为对照,所有样本均设置重复实验。
