We report the use of transcriptome sequencing in the intervention of ANGPTL8 in the regulation of triglyceride level. We constructed the adenovirus-mediated overexpression and knockdown of ANGPTL8 in wild-type mice and obtained the differential genes of ANGPTL8 regulating serum triglyceride level by transcriptomic sequencing. We identified 234 differentially expressed ANGPTL8 response genes, 113 genes are up-regulated in ANGPTL8 overexpression,and down-regulated in ANGPTL8 knock-down. 121 genes are up-regulated in ANGPTL8 down-knock, and down-regulated in ANGPTL8 overexpression. This study provides a framework for the network regulation mechanism of ANGPTL8 controlling triglyceride level. 本研究报道了转录组测序在血管生成素样蛋白8(ANGPTL8)调控甘油三酯水平干预实验中的应用。本研究在野生型小鼠中构建了腺病毒介导的ANGPTL8过表达与敲低模型,并通过转录组测序获取了ANGPTL8调控血清甘油三酯水平的差异表达基因。本研究共鉴定出234个ANGPTL8应答差异表达基因:其中113个基因在ANGPTL8过表达组中上调,在ANGPTL8敲低组中下调;另有121个基因在ANGPTL8敲低组中上调,在ANGPTL8过表达组中下调。本研究为ANGPTL8调控甘油三酯水平的网络调控机制提供了研究框架。整体实验设计:通过RNA测序探究ANGPTL8调控脂质代谢的信号网络。
