Host Factor Experiment (ICL010-P)

Purpose: To obtain samples from Calu-3 cells infected with A/Netherlands/602/2009 (H1N1) and A/CA/04/09 (H1N1) for transcriptional analysis and proteomics analysis for A/Netherlands/602/2009. For A/Netherlands/602/2009 and mock infected the time points are 0, 3, 7, 12, 18, 24, 30, 36 and 48 h post infection. For A/CA/04/09 the time points are 0,12, 24 and 48 h post infection.There are triplicate samples for all conditions for both transcriptional and proteomics analyses. Triplicates are defined as 3 different wells, plated at the same time using the same cell stock for all replicates; Time matched mocks done in triplicate from same cell stock as rest of samples. Inoculation medium for mock infection was the same as the medium used for virus infection. Infection done at an MOI of 3

研究目的：获取感染A/Netherlands/602/2009（H1N1）与A/CA/04/09（H1N1）的Calu-3细胞样本，其中感染A/Netherlands/602/2009的样本同时用于转录组分析与蛋白质组学分析，感染A/CA/04/09的样本仅用于转录组分析。
针对感染A/Netherlands/602/2009的组别及对应时间匹配的模拟感染对照组，取样时间点为感染后0、3、7、12、18、24、30、36及48小时；针对感染A/CA/04/09的组别，取样时间点为感染后0、12、24及48小时。
所有转录组与蛋白质组分析的实验条件均设置3次生物学重复；重复样本定义为铺板时间一致、使用同一细胞储备液的3个独立培养孔；与其余样本来自同一细胞储备液的时间匹配模拟感染对照组同样设置3次重复，其接种培养基与病毒感染组所用培养基完全一致。本实验的病毒感染复数（MOI，Multiplicity of Infection）为3。
