Limited CITE-seq antibody panel and 5' gene expression profiling of human nasopharyngeal cells collected with flocked swabs, paired with TCR and BCR sequencing of NP lymphocytes. FACS-sorted SARS-CoV-2 antigen-specific memory B and T cells with 5' gene expression profiling and scBCR and scTCRseq. Overall design: 10x Genomics 5' immune profiling using BioLegend TotalSeq-C TBNK, hashtag antibodies, and SARS-CoV-2 labeled probes.
本数据集包含两类核心数据:一是采用植绒拭子采集的人鼻咽细胞的有限CITE-seq(Cellular Indexing of Transcriptomes and Epitopes by Sequencing)抗体组合分析与5'端基因表达谱分析,同步配套配对的鼻咽淋巴细胞T细胞受体(TCR)与B细胞受体(BCR)测序;二是经荧光激活细胞分选(Fluorescence-Activated Cell Sorting, FACS)获取的SARS-CoV-2抗原特异性记忆B细胞与T细胞的5'端基因表达谱分析,以及单细胞BCR测序(scBCR)与单细胞TCR测序(scTCRseq)。实验整体设计:采用BioLegend公司的TotalSeq-C TBNK抗体组合、标签抗体及SARS-CoV-2标记探针,通过10x Genomics平台完成5'端免疫组库分析。
