Gene expression in mouse tracheal basal cells

Epithelial basal cells (BCs) are an important stem cell population of the airways. We purified BCs from a KRT5-GFP transgenic mouse line and used Affymetrix arrays to compare there gene expression to that of non-BC epithelium. Three populations of cells were obtained by FACS: 1. KRT5-GFP-, lectin- columnar epithelium. 2. KRT5-GFP-, lectin+ BCs. 3. KRT5-GFP+, lectin+ BCs. Three biological replicates of each sample were hybridized to Affymetrix microarray.

上皮基底细胞（Epithelial basal cells, BCs）是气道中一类重要的干细胞群体。本研究从KRT5-绿色荧光蛋白（GFP）转基因小鼠品系中纯化得到BCs，并利用Affymetrix基因芯片将其基因表达谱与非BC上皮细胞的表达谱进行对比。通过荧光激活细胞分选（FACS）获取了三类细胞群体：1. KRT5-GFP阴性、凝集素（lectin）阴性的柱状上皮细胞；2. KRT5-GFP阴性、凝集素阳性的BCs；3. KRT5-GFP阳性、凝集素阳性的BCs。每个样本设置三次生物学重复，将其与Affymetrix微阵列进行杂交实验。
