Single cell RNA-seq analysis was performed on sorted population of CD45+/CD34+ HSPCs from cord blood (CB) and mobilized peripheral blood (mPB) donors, after co-culture with vascular niche cells, to uncover differential transcriptional output between HSPC derived from CB and mPB origins. Human CB and mPB HSPCs were harvested isolated and sorted. HSPCs were introduced into co-culture with vascular niche ECs. Cultures were harvested and CD45+/CD34+ HSPCs were resorted and processed for 10X Chromium scRNA-seq analysis.
本研究对脐带血(CB)与动员外周血(mPB)供体来源的CD45阳性/CD34阳性造血干祖细胞(Hematopoietic Stem and Progenitor Cells, HSPCs)与血管龛细胞共培养后的群体开展单细胞RNA测序(single cell RNA-seq, scRNA-seq)分析,以揭示两类来源HSPCs的差异转录输出。我们首先从人类CB和mPB供体中收集、分离并分选得到HSPCs,随后将其与血管龛内皮细胞(Endothelial Cells, ECs)进行共培养。培养结束后收集体系,再次分选CD45阳性/CD34阳性的HSPCs,并通过10X Chromium平台完成scRNA-seq分析。
