Early B Cell Factor 2 (EBF2) is required for brown adipose tissue basal thermogenic gene expression; however, it was unknown whether chronic cold exposure normalized the enhancer landscape of Ebf2 mutant mice. We profiled H3K27Ac binding in Ebf2 WT and mutant brown adipose tissue in mice housed at room temperature or one week of cold exposure. H3K27Ac ChIP-Seq was performed in control and Ebf2 mutant brown adipose tissue from adult mice acclimated to room temperature or 1 week of cold exposure.

早期B细胞因子2(Early B Cell Factor 2, EBF2)是小鼠棕色脂肪组织(brown adipose tissue)基础产热基因表达的必需因子;然而目前尚不明确慢性冷暴露是否可使Ebf2突变小鼠的增强子景观(enhancer landscape)恢复正常。本研究采用H3K27Ac染色质免疫共沉淀测序(ChIP-Seq)技术,对饲养于室温环境或经历1周冷暴露的成年小鼠Ebf2野生型(wild type, WT)与突变型棕色脂肪组织的H3K27Ac结合谱型进行了分析。
