Lysine-restricted diet reshapes gut microbes to ease obesity-related anxiety

Total RNA from cortical tissues was extracted and purified using TRIzol reagent (Invitrogen, CA, USA) and DNase I. High-quality RNA samples were used for subsequent analysis. After purification and reverse transcription, RNA transcriptome libraries were constructed using the TruseqTM RNA Sample Preparation Kit and sequenced on the IllumimaNovaSeq6000 platform. In bioinformatics analysis, RSEM was used to quantitatively measure gene abundance. Two groups of differentially expressed genes (DEGs) were identified using DESeq2 .

采用TRIzol试剂（Invitrogen，美国加利福尼亚州）与DNase I对皮层组织的总RNA进行提取与纯化。所获高质量RNA样品用于后续分析。经纯化与逆转录后，使用TruseqTM RNA样本制备试剂盒构建RNA转录组文库，并在Illumina NovaSeq 6000测序平台上完成测序。在生物信息学分析环节，采用RSEM对基因丰度进行定量测定；借助DESeq2鉴定得到两组差异表达基因（differentially expressed genes, DEGs）。
