We previously reported the non-viral derivation of transgene-free induced pluripotent stem cells (iPSCs) from somatic fibroblasts of a female beagle dog using an optimized induction medium and non-transgene-integrating episomal vectors. Here, we report derivation of an induced pluripotent stem cell line OF35Y-iPS from a male beagle dog, which showed standard characteristics of pluripotency such as a strong gene expression profile of pluripotency markers, differentiation potential into all three germ layers, and normal karyotype (78XY). Furthermore, we demonstrated the iPSC line was suitable for genome editing technology. The novel iPSC line would be a useful foothold for stem cell research and regenerative veterinary medicine

我们此前曾报道,通过优化的诱导培养基与无转基因整合的附加体载体(episomal vectors),从雌性比格犬的体细胞成纤维细胞中诱导获得了无转基因整合的诱导多能干细胞(induced pluripotent stem cells, iPSCs)。本研究现报道从雄性比格犬体内诱导建立的诱导多能干细胞系OF35Y-iPS,该细胞系具备多能性的标准特征:多能性标志物的基因表达水平显著,具备向三个胚层分化的潜能,且核型正常(78XY)。进一步实验证实,该iPSC系适用于基因组编辑技术。这款新型诱导多能干细胞系将为干细胞研究与再生兽医学提供宝贵的研究基石。
