We characterized the gene expression profile of brain regions at different stages of the Alzheimer’s like neurodegeneration in the anti-NGF AD11 trangenic mouse model. Total RNA was extracted from hippocampus, cortex and basal forebrain of postnatal day 30 (P30, 1 month), P90 (3 months), P180 (6 months) and at 15 months of age. Expression profiles were studied by Agilent microarray analysis, followed by qRT-PCR validation of significant candidates. Wide changes in gene expression profiles occur already at the presymptomatic stage P30, with the most significantly affected clusters of mRNAs are linked to Inflammation and Immune Response. Overall design: AD11 mice were compared to the trangenic VH controls. A total of 60 female AD11 mice and 59 female control VH (5 per group) mice were used for this study: 5 AD11 and 4-5 control VH mice for each time point (1,3,6,15 months of age) and brain area (HP, CTX, BFB).

本研究对抗神经生长因子(anti-NGF)AD11转基因小鼠模型中类阿尔茨海默病神经退行性病变不同阶段的脑区基因表达谱进行了表征。研究分别选取出生后第30天(P30,1月龄)、第90天(P90,3月龄)、第180天(P180,6月龄)及15月龄的小鼠,提取其海马体(hippocampus)、大脑皮层(cortex)与基底前脑(basal forebrain)中的总RNA。采用安捷伦(Agilent)基因芯片分析对基因表达谱进行检测,并对筛选得到的显著差异候选基因开展qRT-PCR验证。早在症状前阶段P30时,基因表达谱即已出现广泛改变,其中受影响最为显著的mRNA聚类与炎症及免疫应答(Inflammation and Immune Response)相关。 实验设计:将AD11小鼠与转基因VH对照小鼠进行对比。本研究共纳入60只雌性AD11小鼠与59只雌性VH对照小鼠:针对每个时间点(1、3、6、15月龄)及脑区(海马体HP、大脑皮层CTX、基底前脑BFB),每组分别设置5只AD11小鼠与4~5只VH对照小鼠。
