In the present study, we selected 26 clinical isolates of M. tuberculosis carrying a wildtype pncA gene for an in-depth analysis of their phenotypically observed PZA resistance. 7 isolates with confirmed PZA resistance were whole genome sequenced in order to screen for alternative resistance mutations. Our findings may explain most of the phenotypic-genotypic discordances and should be considered when interpreting drug susceptibility test results as well as when evaluating and developing new DST techniques.

本研究选取26株携带野生型pncA基因的结核分枝杆菌（Mycobacterium tuberculosis, M. tuberculosis）临床分离株，针对其表型检测到的吡嗪酰胺（Pyrazinamide, PZA）耐药性展开深入分析。我们对7株经确认具有吡嗪酰胺耐药性的分离株进行全基因组测序，以筛选潜在的耐药突变。本研究结果可解释绝大多数表型-基因型耐药结果不符的情况，在解读药物敏感性试验结果、评估与开发新型药物敏感性试验（Drug Susceptibility Testing, DST）技术时，均应予以参考。
