RNA-seq analysis of mature mouse primary motor cortex in response to conditional knockdown of Fezf2. Analysis of M1 transcriptomes identifies significant changes to gene expression with the knockdown of Fezf2 in mature tissue. Overall design: Lentiviral delivery of a control (non-silencing) or Fezf2 shRNA to mouse primary motor cortex in mature animals (P21-P25; n=4 of each) was used to conditionally reduce Fezf2 expression. Paired-end sequencing was used to generate transcriptome profiles of Fezf2-reduced and control samples.

成年小鼠初级运动皮层响应Fezf2条件性敲低的RNA测序(RNA-seq)分析。对M1区转录组的分析显示,成年组织中Fezf2敲低会导致基因表达发生显著变化。整体实验设计:通过慢病毒载体向成年小鼠(P21-P25日龄,每组样本量n=4)的初级运动皮层递送对照(非沉默型)或Fezf2短发夹RNA(short hairpin RNA, shRNA),以条件性下调Fezf2的表达。采用双端测序技术获取Fezf2表达下调组与对照组样本的转录组图谱。
