Purpose: to characterize the regulatory targets of an AraC-like transcriptional regulator (VC0513) encoded on the Vibrio Seventh Pandemic Island -II (VSP-II) in V. cholerae O1 El Tor N16961 Methods: RNA was isolated from a wild-type N16961 carrying an IPTG-inducible copy of vc0513, vc0515, or an empty vector control Results: vc0513 induction significantly increased expression of other VSP-II encoded genes relative to the empty vector control Conclusions: our study represents the first analysis of a transcriptional regulator encoded on the VSP-II island Overall design: Examination of transcript profiles of vc0513 or vc0515 expressing V. cholerae relative to an empty vector control
研究目的:解析霍乱弧菌O1埃尔特里生物型N16961菌株中,第七次大流行岛-II(Vibrio Seventh Pandemic Island-II,VSP-II)所编码的类AraC转录调控因子(AraC-like transcriptional regulator,VC0513)的调控靶标。实验方法:从携带vc0513、vc0515的IPTG诱导型表达质粒或空载体对照的野生型N16961菌株中提取总RNA。实验结果:与空载体对照组相比,vc0513的诱导表达显著上调了其他VSP-II编码基因的转录水平。研究结论:本研究为针对VSP-II岛编码转录调控因子的首次系统性分析。实验整体设计:对比表达vc0513或vc0515的霍乱弧菌与空载体对照的转录组谱。
