Clr3-CDx2 occupancy along fission yeast chromosomes. Chromodomain-mediated loading targets Clr3-CDx2 to heterochromatin domains. Overall design: DNA co-purified with Clr3-CDx2 chromatin was random-prime PCR amplified and analyzed using custom 60mer Agilent array that tiles Schizosaccharomyces pombe genome in 300bp intervals.
裂殖酵母染色体上Clr3-CDx2的结合占据谱。染色质结构域(Chromodomain)介导的加载过程可将Clr3-CDx2靶向招募至异染色质区域。整体实验设计:与Clr3-CDx2染色质共纯化所得的DNA经随机引物PCR扩增后,采用定制60聚体安捷伦(Agilent)芯片进行检测分析,该芯片以300bp的间隔铺列覆盖粟酒裂殖酵母(Schizosaccharomyces pombe)的全基因组。
