In the present study, we developed a highly accurate Skin-Specific DNAm age predictor, using DNAm data obtained from 508 human skin samples. Based on the analysis of 2,266 CpG sites, we accurately calculated the DNAm age of cultured skin cells, and human skin biopsies. Age estimation was sensitive to the biological age of the donor, cell passage, skin disease status, as well as treatment with senotherapeutic drugs. Analyze changes on the methylation profile of whole skins treated with 100nM Rapamycin compared to no treatment .Sample 1: human whole skin from a 79 year old donor, control, 5 replicates Sample 2: human whole skin from a 79 year old donor, treated with 100nM Rapamycin, 5 replicates

本研究基于508份人类皮肤样本的DNA甲基化(DNA methylation, DNAm)数据,开发了一款高精度的皮肤特异性DNAm年龄预测器。通过对2266个CpG位点的分析,我们可精准计算培养皮肤细胞与人类皮肤活检组织的DNAm年龄。该年龄估算结果对供体生物学年龄、细胞传代代数、皮肤疾病状态,以及衰老治疗药物的干预处理均具有敏感性。本研究同时分析了经100nM雷帕霉素(Rapamycin)处理与未经处理的全层皮肤样本的甲基化谱变化,具体样本信息如下:样本1:来自79岁供体的人类全层皮肤对照样本,设置5次生物学重复;样本2:来自79岁供体的经100nM雷帕霉素处理的人类全层皮肤样本,设置5次生物学重复。
