In vitro propagation of Bucquetia glutinosa, endemic species of the Colombian Paramos

Abstract In vitro multiplication protocol of B. glutinosa was established by seeds collected in the Paramo of Rabanal (Boyacá-Colombia), this species is useful in programs for the conservation and restoration of moorland ecosystems. During the establishment of in vitro cultures, 57% of aseptic seeds were obtained with 5 and 10% NaOCl, after 30 days of culture in MS-1, 61.4% of them developing normal appearance seedlings; 75% of the micro-stems cultivated in MS / 2 + 0.5 mg l-1 IBA reached an average length of 2.06 cm, formed up to 10 axillary buds and among these, around two and four basal shoots were individualized. Root development was parallel to multiplication phase. Plants showed a 94.4% survival rate during acclimation in the greenhouse, on a substrate composed of soil and ground mulch (1:1). This is the first micropropagation protocol of native Colombian melastomáceas with a potential use for restoration and conservation of high Andean ecosystems in a state of deterioration and vulnerability.
摘要 本研究以采自哥伦比亚博亚卡省拉巴纳尔帕拉莫的种子为材料,建立了B. glutinosa的离体繁殖体系。该物种可应用于帕拉莫高寒生态系统的保护与修复项目。在离体培养体系建立过程中,使用5%和10%的次氯酸钠(NaOCl)处理种子,可获得57%的无菌种子;将无菌种子接种于MS-1培养基中培养30天后,其中61.4%可发育为外观正常的幼苗。将微茎段接种于MS/2 + 0.5 mg·l⁻¹吲哚丁酸(IBA)培养基中培养,75%的外植体平均株高达2.06 cm,可形成至多10个腋芽,其中约2~4个基生芽可被单独分离。生根过程与增殖阶段同步进行。组培苗在温室中进行炼苗驯化时,以土壤与地表覆盖物按1:1体积比混合作为栽培基质,最终成活率达94.4%。本研究是首个针对哥伦比亚本土野牡丹科(Melastomataceae)植物的微繁殖方案,可为处于退化与脆弱状态的高海拔安第斯生态系统的保护与修复提供潜在应用价值。
