Our study focuses on understanding how Rbf impacts the formation of flight muscles in Drosophila. We analyzed by RNA-seq the transcriptional changes dependent on Rbf in proliferating adult muscle precursors and in mature flight muscles at two different stages of Drosophila development Overall design: Total RNA was extracted at two different developmental stages: notum tissues from third instar larval wing discs, which contains adult muscle precursors, and the flight muscles of pharate flies. Total RNA libraries were made and followed by sequencing. The following two genotypes were compared at these two developmental stages: WT and Rbf-conditional knockdown using Mef2-Gal4 driver and UAS-Rbf-RNAi. Three replicates were done for notum tissue and two replicates for flight muscles
本研究旨在阐明Rbf对果蝇(Drosophila)飞行肌形成的调控机制。我们通过RNA测序(RNA-seq)分析了果蝇发育两个不同阶段中,依赖于Rbf的增殖期成体肌前体细胞以及成熟飞行肌的转录组变化。实验设计概述:我们于两个不同发育阶段提取总RNA:其一为含有成体肌前体细胞的三龄幼虫翅盘背甲组织,其二为即将羽化果蝇的飞行肌。随后构建总RNA文库并完成测序。本实验在上述两个发育阶段中比较了两种基因型样本:野生型(WT)以及借助Mef2-Gal4驱动元件与UAS-Rbf-RNAi实现的Rbf条件性敲低组。其中背甲组织设置3次生物学重复,飞行肌设置2次生物学重复。
