We have identified HNF6 as an essential transcription factor for endocrine pancreatic development. HNF6 binds to and possibly regulates other isletogenesis transcription factors, driving endocrine differentiation. ChIP sequencing of HNF6 as well as GATA6 and NKX6.1 during in vitro pancreatic differentiation will help to understand the regulatory mechanisms during cell fate decisions. The human embryonic stem cell line CyT49 differentiated to gut tube endoderm and pancreatic progenitor cells were used for chromatin immunoprecipitation.

本研究已证实HNF6是胰腺内分泌发育过程中的关键转录因子。HNF6能够结合并可能调控其他胰岛生成相关转录因子,从而驱动内分泌细胞分化。对体外胰腺分化过程中的HNF6、GATA6及NKX6.1进行染色质免疫共沉淀测序(ChIP sequencing),将有助于阐明细胞命运决定过程中的调控机制。本研究采用经诱导分化为肠管内胚层与胰腺祖细胞的人类胚胎干细胞系CyT49开展染色质免疫沉淀实验。
