Methylation array was performed to understand the role of UHRF1 in DNA methylation in KRAS mutant lung cancer cell lines. Knock-down of UHRF1 leads to significant changes in global DNA methylation in both cell lines. Two KRAS mutant lung cancer cell lines were used (A549, H358). Each cell line was treated with control siRNA (siNeg), anti-KRAS siRNA (siKRAS) or anti-UHRF1 siRNA (siUHRF1) in 2 (siNeg) or 3 (siUHRF1, siKRAS) biological replicates.
本研究采用甲基化芯片(methylation array)技术,旨在阐明UHRF1在KRAS突变型肺癌细胞系DNA甲基化调控中的功能。敲低UHRF1可使两种细胞系的全基因组DNA甲基化水平发生显著变化。本研究所用的两种KRAS突变型肺癌细胞系为A549与H358。针对每种细胞系,分别设置对照小干扰RNA(siNeg)、抗KRAS小干扰RNA(siKRAS)及抗UHRF1小干扰RNA(siUHRF1)三种处理组;其中siNeg组设置2次生物学重复,siUHRF1与siKRAS组各设置3次生物学重复。
