From this array we would like to know the role of Smyd2 on hypoxic regulated genes mainly angiogenic pathway. We performed overexpressions of Smyd2 and beta galactosidase by adenoviral approach and knockdown of HIF1 and Smyd2 were performed by lipofectamine RNAiMAX.Cardiomyocytes were isolated from neonatal rats at P3. 2 groups, each with 2 biological replicates
基于本基因芯片实验,我们旨在探究Smyd2在缺氧调控基因(主要涉及血管生成通路)中的调控作用。本研究通过腺病毒介导的方式完成Smyd2与β-半乳糖苷酶的过表达,并借助Lipofectamine RNAiMAX试剂实现缺氧诱导因子1(HIF1)与Smyd2的基因敲低。实验所用心肌细胞分离自出生后第3天(P3)的新生大鼠,共设置2组,每组均包含2个生物学重复样本。
