We generated time-resolved profiles of fasting-induced changes in the genomic landscapes of hepatocytes and liver macrophages using the INTACT methodology We subjected female HEP-INTACT and MAC-INTACT mice to a fast starting at the onset of the mouse active phase, ZT12, and after 3, 8, 16, and 24h livers were collected for INTACT followed by ATAC-seq analyses. Ad libitum fed controls were sacrificed at each time point to account for diurnal variations in gene expression.
本研究采用特异性细胞类型标记细胞核分离技术(INTACT),构建了禁食诱导下肝细胞与肝巨噬细胞基因组景观动态变化的高时间分辨率图谱。我们让雌性HEP-INTACT与MAC-INTACT小鼠于小鼠活动期起始时刻ZT12开始禁食,并分别于禁食3、8、16及24小时后采集肝脏样本,经INTACT分选后开展转座酶可及性染色质测序(ATAC-seq)分析。为校正基因表达的昼夜节律波动,我们在每个采样时间点均设置自由进食对照组,并对其实施安乐死。
