Stevia rebaudiana is an important medical plant for producing steviol glycosides (SGs) or stevioside. Autotetraploids (4x = 44) show an increasing level of morphology, physiology and tolerances comparing to diploids (2x =22). By constucting a comprehensive transcriptome of stevia leaf, stem and root from the diploids and autotetraploids based on RNA-seq, we want to obtain a primary transcriptome assembly of S.rebaudiana and identify differentially expression genes between autotetraploids and diploids, advancing our understanding in molecular changes related to SGs metabolism of polyploidy, plant resistance responsible for phenotypic change of autotetraploids.

甜叶菊(Stevia rebaudiana)是用于生产甜菊糖苷(steviol glycosides, SGs)或甜菊苷的重要药用植物。相较于二倍体(diploids, 2x=22),同源四倍体(autotetraploids, 4x=44)在形态学、生理学特性与抗逆性水平上均表现更优。本研究基于RNA测序(RNA-seq)技术,对二倍体与同源四倍体甜叶菊的叶片、茎秆及根系开展全面转录组(transcriptome)构建工作,旨在获得甜叶菊的初步转录组组装结果,并鉴定同源四倍体与二倍体间的差异表达基因,从而助力我们深入理解多倍体(polyploidy)甜菊糖苷代谢相关的分子变化,以及调控同源四倍体表型改变的植物抗性机制。
