We found that targeting IDH1 in HGSC induces senescence by altering the repressive histone landscape of E2F target genes Overall design: Ovcar3 cells were infected with control or shIDH1. Cells were selected in 3ug/mL puromycin for 7 days. RNA was isolated and RNA-seq was performed comparing control and knockdown RNA.
本研究发现,在高级别浆液性卵巢癌(High-grade Serous Ovarian Carcinoma, HGSC)细胞中靶向异柠檬酸脱氢酶1(Isocitrate dehydrogenase 1, IDH1),可通过改变E2F靶基因的抑制性组蛋白调控图谱,诱导细胞衰老。实验设计概述:将Ovcar3细胞分别用对照慢病毒及靶向IDH1的短发夹RNA(shIDH1)慢病毒感染,随后以3 μg/mL嘌呤霉素筛选7天。提取各组细胞总RNA,对对照组与IDH1敲低组的RNA进行RNA测序(RNA-seq)以开展对比分析。
