Osteopontin expression identifies a subset of recruited macrophages distinct from Kupffer cells in the fatty liver. [Bulk RNA Seq]

CITE-seq analysis on total CD45+ cells isolated from the livers of mice fed a standard diet (SD) or western diet (WD; fat, cholesterol and sugar) for 12, 24 and 36 weeks. We also performed single cell RNA sequencing analysis of the CD45- cells isolated from the livers of the mice fed the SD or WD for 24 and 36 weeks. Single cell RNA Seq = Liver CD45+ and CD45- cells derived from mice fed a standard diet (SD) or western diet (WD; fat, cholesterol and sugar) for 12, 24 and 36 weeks. Bulk RNA Seq = Kupffer cells, moKCs and Clec4f- macrophages derived from mice fed a standard diet (SD) or western diet (WD; fat, cholesterol and sugar) for 12 and 24 weeks.本研究针对喂食标准饲料（SD）或西式饲料（WD，含脂肪、胆固醇与蔗糖）12、24、36周的小鼠肝脏中分离得到的总CD45阳性细胞，开展了细胞转录组与表位联合测序（CITE-seq）分析。同时，本研究还对喂食SD或WD 24、36周的小鼠肝脏分离的CD45阴性细胞，进行了单细胞RNA测序 (single cell RNA sequencing, scRNA-seq) 分析。其中，单细胞RNA测序的全部样本范围为：喂食标准饲料（SD）或西式饲料（WD，含脂肪、胆固醇与蔗糖）12、24、36周的小鼠肝脏来源的CD45阳性与CD45阴性细胞。批量RNA测序 (Bulk RNA Seq) 的样本为：喂食标准饲料（SD）或西式饲料（WD，含脂肪、胆固醇与蔗糖）12、24周的小鼠肝脏来源的库普弗细胞 (Kupffer cells)、单核细胞源性库普弗细胞 (moKCs) 以及Clec4f阴性巨噬细胞。
