Here we found that Cbx8 knockdown in MMTV-Myc and MDA-MB-231-Luc cells reduces H3K4me3 mark on Notch-network genes Overall design: Mononucleosomes from MMTV-Myc and MDA-MB-231-Luc cells were isolated and ChIP with H3K4me3, H3K27me3 and H3K27ac antibodies. DNA from Input and ChIP samples was purified and sequenced on Illumina Hiseq.

本研究发现,在MMTV-Myc细胞及MDA-MB-231-Luc细胞中敲低染色质盒同源物8(Cbx8),可降低Notch网络基因上的组蛋白H3K4三甲基化(H3K4me3)修饰水平。实验设计:分离MMTV-Myc与MDA-MB-231-Luc细胞的单核小体,使用针对H3K4me3、H3K27me3及H3K27ac的抗体开展染色质免疫共沉淀(Chromatin Immunoprecipitation, ChIP)实验;对输入对照(Input)样本及ChIP富集样本的DNA进行纯化后,采用Illumina HiSeq测序平台完成测序。
